Strain Description
This mouse strain is constructed by placing the optimized iCre recombinase gene under the control of the Villin 1 (Vil1) gene promoter/enhancer, generating a transgenic mouse line. Vil1 is a cytoskeletal protein that is widely and specifically expressed in intestinal epithelial cells (from the small intestine to the colon). Its promoter drives gene expression in virtually all intestinal epithelial cell types, including stem cells at the base of the crypts, proliferating progenitor cells, differentiated absorptive cells, goblet cells, enteroendocrine cells, and Paneth cells. This model drives efficient and specific expression of iCre recombinase in these Vil1-positive cells (i.e., intestinal epithelial cells). When crossed with reporter mice carrying LoxP sites (e.g., Rosa26-LSL-tdTomato) or conditionally modified mice (Floxed mice), it enables stable reporter gene expression or conditional gene knockout/activation throughout the intestinal epithelial lineage. This model is one of the most important and commonly used genetic tools for studying intestinal development, physiology, stem cell biology, intestinal diseases (such as IBD and colon cancer), and host-microbiota interactions.
